Riboflavin 5′-Phosphate Sodium

Riboflavin 5′-phosphate sodium (CAS 130-40-5), also called riboflavin sodium phosphate or FMN sodium, is the water-soluble form of vitamin B2 used in liquid supplements, beverages and other clear formulations.

Identity

What it is.

Riboflavin itself barely dissolves in water. Adding a phosphate group and forming the sodium salt gives a compound that dissolves readily, which is why riboflavin 5′-phosphate sodium is the form used in liquid vitamins, syrups, drops, shots and clear beverages.

Its assay is expressed differently from riboflavin. The USP monograph specifies the content as riboflavin, at 73.0% to 79.0% on the dried basis, because part of the molecule's weight is the phosphate and sodium. Our material is produced to that monograph; a recent batch assayed at 74.8%.

Also known as: Riboflavin sodium phosphate, Riboflavin-5′-phosphate sodium, FMN sodium, Flavin mononucleotide sodium, E101(ii).

Questions

Riboflavin 5′-Phosphate Sodium: common questions.

What is riboflavin 5′-phosphate sodium?

It is the sodium salt of riboflavin 5′-phosphate, also known as flavin mononucleotide (FMN). It delivers vitamin B2 in a form that dissolves in water. Its CAS number is 130-40-5.

What is the difference between riboflavin and riboflavin 5′-phosphate sodium?

Both supply vitamin B2. Riboflavin is very slightly soluble in water and assays at 98% or more. Riboflavin 5′-phosphate sodium is soluble in water and contains 73% to 79% riboflavin by weight, so more of it is needed to deliver the same amount of vitamin B2. Dry products usually use riboflavin; liquids usually use the phosphate sodium salt.

Why is the assay 73% to 79% and not 98%?

The USP assay for riboflavin 5′-phosphate sodium is reported as riboflavin. The phosphate group, the sodium and the water of hydration make up the rest of the weight. A lot that meets the monograph is fully in specification at about 75%.

How much riboflavin 5′-phosphate sodium replaces 1 mg of riboflavin?

Divide by the assay. At a typical 75% riboflavin content, about 1.33 mg of riboflavin 5′-phosphate sodium supplies 1 mg of riboflavin. Use the assay on the certificate of analysis for the lot you are formulating with.

Is riboflavin 5′-phosphate sodium GRAS?

Yes. It is affirmed as GRAS for use as a nutrient supplement in food under 21 CFR 184.1697.
